Imaging signal transduction in single dendritic spines  

Many forms of mental retardation and autism are caused by abnormal intracellular signaling cascades involving proteins in the Rho family. The shape and function of synapses are regulated by signaling mediated by Rho proteins. This project will develop a novel cellular imaging technique to measure the activity of Rho proteins in single synapses, leading to a fuller understanding of how Rho activity helps to control the function of synapses. This study will illuminate some of the molecular mechanisms of synaptic plasticity and will provide insights into diseases caused by mutations in Rho signaling pathways, such as mental retardation and autism.
